Product Introduction
Human milk oligosaccharides (HMOs) are complex carbohydrates naturally occurring in human breast milk and are the third largest solid component in breast milk (after lactose and fat), which are composed of 5 basic simple sugars through various linkages, with more than 200 structures identified. This active ingredient, known as the "soft gold" of breast milk, has achieved commercial application in infant nutrition, health care and other fields with its unique physiological functions, and has become the core direction of simulated breast milk formulation.
The core characteristics of breast milk oligosaccharides are reflected in "breast milk homology", "structural diversity" and "biological activity", which can be deeply understood from three aspects: source process, physical and chemical characteristics, and core specifications:
1. Source and production process
The content of HMOs in natural breast milk is extremely low and difficult to extract, and commercial products mainly rely on artificial synthesis, and the mainstream process has been upgraded from chemical synthesis to efficient biosynthesis:
Natural source: found only in human breast milk, highest in colostrum (average 9-22g/L) and gradually decreased in mature milk with prolonged lactation (4-6g/L after 6 months). The composition of HMOs in different breast milk is affected by maternal genotype, mode of delivery, etc., for example, the content of 6'-sialic acid lactose (6'-SL) in colostrum of a vaginal mother is higher than that of a cesarean section mother.
Commercial production: At present, microbial fermentation is the mainstream, using lactose and glucose extracted from plant starch as raw materials, and targeted synthesis of target HMOs by genetically engineered microorganisms (such as Escherichia coli and yeast), which are purified and spray-dried into powder products. This process allows for the precise production of HMOs with specific structures (e.g., 2'-FL) with high purity and consistent structure with natural breast milk ingredients, and has passed food safety certifications such as FSSC 22000. In addition, there are auxiliary processes such as enzymatic method and chemical enzymatic method to meet different purity requirements.
2. Core physical and chemical properties
The physical and chemical properties of HMOs are directly related to their biological functions and application scenarios, and the core features are as follows:
Solubility and stability: easily soluble in water, can be quickly dissolved in liquid matrix after being made into powder, and the solution is clear and non-precipitated; It has good stability in neutral and slightly acidic environments (pH 4.0-7.0) and room temperature, and can withstand conventional processing processes (such as pasteurization) of infant formula food, but long-term treatment at high temperature (>140°C) will lead to partial structural degradation.
Digestive properties: As "indigestible carbohydrates", HMOs function intact through the upper digestive tract to the distal small and large intestines, and are not absorbed by the small intestine, resulting in virtually no heat production and no blood sugar fluctuations.
Structural diversity: Based on glycan modification characteristics, it can be divided into three categories - neutral fucosylation (e.g., 2'-FL), sialylation (e.g., 3'-SL, 6'-SL), and neutral non-fucosylation (e.g., LNT), with different structures corresponding to different biological functions.
3. Core specifications and quality standards
Commercialized products exist as a single ingredient or in compounds, with core indicators of "structural purity" and "microbiological safety":
Mainstream single product specifications: mainly high-purity powders with a content of ≥ 90%, common varieties include 2'-fucosyllactose (2'-FL, the most abundant HMOs in breast milk), lactose-N-neotetrasaccharide (LNnT), 3'-sialic acid lactose (3'-SL), etc.
Compound specifications: According to the characteristics of breast milk ingredients, 2'-FL is compounded with LNnT, 3'-SL, etc., to simulate the composition ratio of HMOs in natural breast milk to meet the needs of infant formula food.
Safety standards: It is necessary to comply with food safety regulations of various countries, such as the US FDA listing Class 8 HMOs as GRAS (generally recognized as safe) substances, and China has clear limit requirements for the addition of HMOs in infant formula.
Application of the product
Breast milk oligosaccharides have achieved precise applications in infant nutrition, health food, special medical uses, etc.
1. Infant formula (core application areas)
At the heart of this application is "simulated breast milk function", which has become a signature ingredient in high-end formulas:
Formula milk powder: Add single or compound HMOs such as 2'-FL and LNnT for infant formula and special formula for premature infants, claiming "breast milk-grade intestinal protection" and "brain development" and other selling points. The amount added is usually 0.3-2.0g/L, which is close to the natural content in breast milk. Formulas containing 2'-FL and LNnT have been shown to bring infant gut microbiota composition closer to that of breastfed infants.
Complementary foods and nutritional products: used in infants and young children's rice noodles, fruit purees, nutritional supplements (such as drops), especially suitable for infants and young children with high risk of allergies and sensitive intestines. For example, adding sialylated HMOs to complementary foods can provide both gut protection and brain developmental support.
2. Health food and health products
For the health needs of adults and special populations, the application of HMOs has expanded from infants and young children to a wider range of fields:
Gut health products: Compounded with probiotics and dietary fiber to make solid drinks and capsules, focusing on "regulating intestinal flora" and "improving irritable bowel syndrome (IBS)", suitable for office people, the elderly and other intestinal dysfunction. Clinical studies have shown that HMOs can significantly increase the proportion of beneficial bacteria in IBS patients, relieving symptoms such as bloating and diarrhea.
Immune support products: For people with weakened immunity (such as those recovering from surgery and the elderly), oral liquids or tablets are made to strengthen mucosal immunity and modulate immune responses. Some products combine vitamin C, zinc and other ingredients to achieve "multiple immune protection".
Brain health food: For children, adolescents and the elderly, cereal breakfasts and nutritional bars with sialylated HMOs (such as 6'-SL) claim to "promote neurodevelopment" and "maintain cognitive function", and provide gentle support with the help of the "gut-brain axis" mechanism.
3. Food for special medical purposes
In the field of medical nutrition, HMOs are used for disease-specific adjunctive interventions due to their precise biological functions:
Nutrition for premature infants: It is added to enteral nutrition preparations for premature infants to reduce the risk of necrotizing enterocolitis, infectious diseases, and promote intestinal maturation.
Metabolic disease assistance: For people at high risk of type 1 diabetes, relevant studies have shown that HMOs can inhibit the development of pancreatitis by modulating the intestinal flora, and are expected to be used in diabetes prevention medical foods in the future.
Allergy management: Add it to the special diet of patients with allergic diseases to reduce inflammatory responses by regulating immune balance and assist in improving symptoms such as eczema and allergic rhinitis.
4. Maternal and child care
With their mild antibacterial properties, HMOs have been extended to topical care products for mother and baby:
Infant skin care: Add to buttock balm and wipes, by inhibiting the growth of pathogenic bacteria such as Staphylococcus aureus and Escherichia coli, reduce the risk of red buttocks and skin infection, and the pH value is close to that of infant skin (5.5-6.5), without irritation.
Oral care: Used in oral cleaning solution for infants and young children, inhibiting the colonization of oral caries-causing bacteria, while providing gentle protection for the gingival mucosa and preventing early caries.
