Applications of Lactulose in Medical Health Industry
What is LOSWIFT™Lactulose?
Natural Sweetness, Enhanced Performance
LOSWIFT™Lactulose (4-O-beta-D-galactopyranosyl-D-fructose) is a synthetic disaccharide in which a galactose unit is joined to a fructose unit through a beta-1,4 glycosidic bond. It is not found in nature and is commercially manufactured by alkaline isomerization of lactose, the milk sugar, via the Lobry de Bruyn-Alberda van Ekenstein rearrangement. With a molar mass of 342.30 g/mol and a melting point around 169 degrees C, LOSWIFT™Lactulose is a white, odourless crystalline powder with a mild sweet taste.
Because human digestive enzymes cannot hydrolyse its glycosidic linkage, LOSWIFT™Lactulose passes intact through the upper gastrointestinal tract and reaches the colon, where it acts as a selective substrate for beneficial saccharolytic bacteria. This property underpins its long-standing role as a prebiotic and pharmaceutical agent, registered as a medicinal drug in more than 100 countries for indications including constipation and hepatic encephalopathy.
In recent years, its application scope has broadened to encompass functional foods, clinical nutrition, and feed additives.
Health Benefits
Relief and Long-Term Management of Constipation
LOSWIFT™Lactulose acts as an osmotic laxative approved across all life stages. Fermented by caecal flora into short-chain fatty acids, it lowers luminal pH, increases osmotic load to draw water into the colonic lumen, enlarges stool bulk, and accelerates propulsive motility without triggering cramping or disturbing electrolyte balance.
Lowering of Blood Ammonia and Support in Hepatic Encephalopathy
Functions as a cornerstone first-line treatment for portal systemic encephalopathy. Acidification converts diffusible NH3 into non-diffusible NH4+ lost in faeces, while prebiotic expansion of microbial biomass channels nitrogen into protein synthesis and restrains ammonia-generating bacterial enzymes.
Suppression of Secondary Bile Acid Formation
Selectively enriches saccharolytic organisms to acidify the colonic milieu, bringing a clear fall in both faecal concentration and daily excretion of cytotoxic secondary bile salts (lithocholic and deoxycholic acids) to help normalize biliary acid balance.
Anti-Endotoxin Effects in Obstructive Jaundice
Exhibits anti-endotoxin activity in surgical contexts by lowering systemic blood endotoxin levels and mitigating postoperative endotoxaemia, protecting against endotoxin-mediated complications such as renal dysfunction.
Possible Protective Roles in Colon Carcinogenesis and Tumour Defence
Reduces exposure to tumour-promoting secondary bile salts and suppresses colonic mucosal DNA damage, while fostering bifidobacteria to trigger immune-enhancing and potential chemopreventive responses for long-term colon health support.
Application Advantages
Value in Managing Nontyphoid Salmonella Carriers
Provides a non-antibiotic route to accelerate Salmonella elimination. High doses induce pronounced colonic acidification that renders the environment inhospitable to persistent Salmonella, achieving faster clearance rates than non-treatment or antibiotic regimens.
Adjunctive Potential in Inflammatory Bowel Disease
Acts on both microbiota and bacterial endotoxins by raising organic acid production and lowering faecal pH to favor L. acidophilus while restraining pathogens. Regularizes bowel habits, acts as an indirect antioxidant via colonic hydrogen generation, and helps maintain remission in IBD.
Promotion of Mineral Absorption and Bone Health
Markedly enhances intestinal uptake and retention of essential minerals, above all calcium and magnesium (as well as zinc, copper, and iron). Colonic acidification solubilises mineral salts while bacterial fermentation upregulates mucosal transport pathways to improve bone mineralization.
